The Ulanbuhe Desert is located in the western part of the Inner Mongolia Autonomous Region, with a total area of 15 million acres. The sand area is rich in animal and plant resources, and the sand industry has a long reputation. Among them, the traditional Chinese patent medicines and simple preparations Cistanche deserticola, which has the reputation of "desert ginseng", is famous at home and abroad. The Ulanbuhe Sand District has advantageous conditions for the development of the Cistanche industry, and after more than ten years of development, the Cistanche industry has begun to take shape. However, several technological bottlenecks that have hindered the development of the Cistanche deserticola industry have not been properly addressed, directly limiting its sustainable development. In response to a series of issues such as low planting efficiency, high labor consumption, and high planting costs in the current cultivation process of Cistanche deserticola ma, the focus is on researching the inoculation technology of Cistanche deserticola, continuously expanding the development of the Cistanche deserticola industry, and playing an important role in controlling the ecological environment in sandy areas, improving people's livelihoods, and achieving sustainable development.

1、 Inoculation techniques for Cistanche deserticola

Cistanche deserticola can be inoculated using two technical modes: mechanical integration inoculation, manual drilling and digging, and trenching inoculation (mainly using sand seed mixing, inoculation paper, and inoculation block).

1. Mechanically integrated trenching and inoculation

According to the inoculation amount of Cistanche deserticola seeds, 10 grams, 20 grams, and 30 grams per acre were mechanically inoculated. Use a Cistanche seeding machine to open a ditch at a distance of 20-40 centimeters beside the shuttle, with a width of 30 centimeters and a depth of 40-80 centimeters, to sow Cistanche seeds and organic fertilizers. The vertical spiral seeder driven by the ground wheel can accurately control the sowing amount of Cistanche. Through multiple layers of seeding pipes and reflective plates, the Cistanche seeds are evenly spread in a three-dimensional space of 40-80 centimeters underground; At the same time, a horizontal spiral fertilizer feeder driven by land wheels is used to accurately control the fertilization amount, and the fertilizer is evenly spread within the range of 20-30 centimeters through a fertilizer pipe.

2. Artificial vaccination

(1) Digging and Trenching

Dig a planting pit with a depth of 70-100 centimeters, a length of 80-100 centimeters, and a width of 50-80 centimeters at a distance of 20-40 centimeters from the Haloxylon ammo dendron plant. The shorter the vaccination distance, the higher the success rate of vaccination, and the shorter the time for successful parasitism. But being too close can easily cause artificial damage to the branches and main root system of Haloxylon ammo dendron; After the excavation of Cistanche deserticola, it can be inserted into the gaps in the branches of Haloxylon ammo dendron, making it difficult to collect seeds or excavate Cistanche deserticola. The depth of trenching is related to the length, thickness, and yield of the stem of Cistanche deserticola.

(2) Mixed sand seed preparation

The seeds of Cistanche deserticola have fine particles, about 9000 seeds per gram. It is not easy to control the sowing amount by manual sowing, and it needs to be mixed with about 100 times the amount of wet yellow sand. Calculate the sowing amount per mu of Cistanche deserticola seeds in advance, accurately weigh them, and then mix them thoroughly with sand with a humidity of about 65% to prepare mixed sand seeds for sowing.

(3) Sowing amount

The trenching inoculation method has a large sowing volume and a three-dimensional distribution of seeds, which is also an effective way to achieve a high success rate of inoculation. If the seed maturity is good, apply 10-50 grams per acre, with an average of 20 grams.

(4) Sowing

When opening the ditch, evenly place the excavated sand on both sides of the ditch mouth, and evenly spread the mixed sand seeds on the inclined surface of the sand ridge near the host seedling. Inoculation paper and blocks can also be placed on the side near the plant. Seeds cannot be directly sown into the ditch, otherwise, the three-dimensional sowing layer will be too thin and not achieve good results.

(5) Seed entering groove

Before inoculation, place some fermented sheep manure in the pit appropriately. The construction personnel enters the ditch, facing the host shuttle with their backs, using both hands to mix in layers from bottom to top, and plowing sand and soil into the ditch. The thickness of the seed layer in the furrow should be maintained at 10-15 centimeters, which can be completed in three steps. First, stir and scrape into the upper one-third section of the covered sand ridge, then the middle section, and finally the lower one-third section. Put all the seeds on the ridge surface into the ditch, and the mixed sand seed layer inside the ditch should be evenly and flat. At this point, the seed layer should be about 60 centimeters away from the ground surface. Finally, backfill 30 centimeters of sand and cover the seed layer to prevent water seepage during watering and prevent the seeds from drifting and floating due to water flow.

After sowing, reserve a watering ditch of about 25 centimeters at the mouth of the ditch, and use the water pump and a water pipe with a diameter of 1.5 inches or more in the forest to water or drip until it has fully penetrated. Then fill and compact all the sand and soil at the mouth of the ditch, and cover it with a thin layer of sand about 5 centimeters to retain moisture.

2、 Nursing management and comparative analysis

A survey was conducted on the survival rate of Haloxylon ammo dendron in August 2020, and in October 2022, a control experiment was conducted in different areas based on different irrigation methods, inoculation amount of Cistanche deserticola seeds, and inoculation frequency.

1. Survival of Haloxylon ammo dendron under different irrigation methods

The survival rates of Haloxylon ammo dendron in drip irrigation and traditional irrigation methods were investigated separately (Table 1). The survival rates in drip irrigation areas reached 91%, while those in traditional irrigation areas reached 84%. The survival rates of Haloxylon ammo dendron in drip irrigation areas were higher than those in traditional irrigation areas. The use of drip irrigation is more water-saving than traditional irrigation methods, and the cost per acre is more than 150 yuan lower than traditional irrigation.

2. Investigation on the yield of Cistanche deserticola with different inoculation doses

By using mechanical inoculation, in October 2022, sample plants were selected for investigation from plots with the same irrigation frequency (4 times) and different inoculation amounts (10 grams, 20 grams, and 30 grams of Cistanche deserticola seeds per acre). The results showed that with an inoculation amount of 10 grams per mu, the average yield of Cistanche deserticola per plant was 0.79 kilograms, and the yield per mu was 88 kilograms; The inoculation amount per mu is 20 grams, and the average yield of Cistanche deserticola is 0.98 kilograms per plant, with a yield of 109 kilograms per mu; The inoculation amount per mu is 30 grams, and the average yield of Cistanche deserticola per plant is 1.36 kilograms, with a yield of 151 kilograms per mu. Through comparison, it can be concluded that after inoculating 30 grams of Cistanche deserticola seeds, the yield per mu of Cistanche deserticola is significantly higher than that of 10 grams and 20 grams.

3. Investigation on the Yield of Cistanche deserticola with Different Drip Irrigation Times

Using mechanical integration inoculation, with an inoculation amount of 30 grams, the optimal drip irrigation frequency was studied based on 3, 4, and 5 different drip irrigation times to improve water-saving efficiency. Sample plants were selected in October 2022 to investigate the yield of Cistanche deserticola (Table 3). The results showed that after three rounds of irrigation, the average yield of Cistanche deserticola sample plants was 0.93 kilograms, and the yield per mu was 103 kilograms; Irrigation 4 times, the average yield of Cistanche deserticola sample plants is 1.36 kilograms, and the yield per mu is 151 kilograms; Irrigation 5 times, the average yield of the Cistanche deserticola sample plant is 1.41 kilograms, and the yield per mu is 157 kilograms. By comparison, it can be concluded that the highest yield occurs when irrigated 5 times.

3、 Conclusion

By adopting mechanical integration inoculation, under drip irrigation, the survival rate of Haloxylon ammo dendron was higher than that of traditional irrigation. The highest yield of Cistanche deserticola was obtained when the inoculation amount was 30 grams and irrigation was conducted 5 times.

By solving these technical problems, it is possible to scientifically and reasonably develop and sustainably utilize the resources of Cistanche deserticola, which also plays an important role in the healthy development of society, ecology, and economy in the Ulanbuhe sandy area.
