Laxative Action Of Separated Parts From Herba Cistanches

Abstract Objective: To search the herba cistanches for effective corporation on laxatives.
Methods: The herba cistanches are divided into galactitol and others through methods of extraction and separation and crystal with different solvents. The four sections are tested by the time of pass through the stomach and intestines of mice.
Results: Galactitol can obviously shorten the time of pass through the stomach and intestines, the action of others is not obvious.
Conclusion: Galactitol is the main effective corporation on laxatives.

The clinical practice of traditional Chinese medicine proves that Cistanche has a good function of moisturizing the bowel and laxative. The previous articles [1, 2] have reported the laxative function of the drug and the modern pharmacological experimental study of the relevant mechanism of action. This article reports the preliminary extraction and separation of various parts of Cistanche and the experimental results of the sample liquid for each part to be used for a laxative.
1. Experimental materials
1.1 Medicinal materials
The original medicinal material of Cistanche was purchased from Alexa League in Inner Mongolia. It is a super product. It was identified as Cistanche deserticoleY.C.Ma by Tu Pengfei from the School of Pharmacy of Peking University, also known as Cistanche deserticola.
1.2 Test solution
Take the original medicinal materials, dry them at low temperature, crush them into a coarse powder, and extract and separate them according to the process shown in Figure 1 to obtain galactitol crystals, ethyl acetate extract, n-butanol extract, and remaining water-soluble substances. Before the experiment, distilled water was used to prepare the test solution (containing 1% CMC-Na and 1% eosin). The concentration of the drug solution except for β-galactitol, and the extracted content in the remaining test solutions are equivalent to the amount of the original medicinal material 100%.

2. Methods and results
2.1 The effect on mice's laxative time
Sixty healthy mice weighing 18-20g were selected and randomly divided into 5 groups according to their body weight, with 12 mice in each group. Fasting and water-removing 2h before administration, the mice were placed in single breeding cages, and each group was given 0.5mL/mouse for the test solution, and the administration time and the time for the mice to discharge the first red feces were recorded, and the latter was subtracted. The former is the laxative time of the rat. A total of 2 batches of experiments were done, the results are shown in Table 1 and Table 2.

The results of the two batches showed that the isolated component of Cistanche, galactitol, had a significant effect on shortening the laxative time, and the effect of the other groups was not significant.
2.2 The effect of doubling the dose on the time of defecation in mice
In the above experiment, except for the separated component galactitol, the component without significant laxative effect doubled the dose and then carried out the experiment on the effect of the mouse laxative time, the method is the same as above. There are two batches of experiments, and the results are shown in Table 3 and Table 4. The experimental results show that even though the dose of n-butanol extract and other components is increased by 1 time, there is no significant effect of shortening the laxative time.

3. Conclusion
The experimental results show that among the four parts of the extracts initially separated from Cistanche, only the alcohol-soluble part of galactitol has a definite laxative effect, and the other extracts have no obvious laxative effect. The second batch of experiments confirmed the results of the first batch of experiments. On the basis of the two batches of experiments, we doubled the dose of ethyl acetate extract, n-butanol extract, and the remaining water-soluble part, and performed a laxative experiment again. We still did not see that these extracts have a laxative effect. The above experimental results indicate that the galactitol contained in Cistanche is the main laxative active ingredient of Cistanche. The inorganic ions such as sodium, magnesium, sulfate, etc. reported in previous articles may only play an auxiliary role in the overall laxative effect of Cistanche.
