Cistanche Extract Supplements For Men's Health Improvement

Description

Technical Field

[0001] The present invention relates to a ginseng–Cistanche–Polygonatum composition for improving erectile dysfunction caused by aging and a preparation method thereof.

Background Art

[0002] Ginseng and Polygonatum cultivated for five years or less are materials used both as food and medicine. Their traditional processing methods involve one or multiple cycles of steaming and sun‑drying (Polygonatum traditionally undergoes "nine steaming and nine drying"), and the processed products are referred to as red ginseng, "Xianren Yuliang", etc., and are considered to have anti‑aging effects and to promote male reproductive health. The food‑and‑medicine material Cistanche is considered to improve male health and soften blood vessels.

[0003] Traditionally, the beneficial health effects of red ginseng are attributed to low‑polarity saponins such as Rg2, Rh1, and Rg3 and to ginseng polysaccharides; the effects of "Xianren Yuliang" are usually attributed to Polygonatum polysaccharides, flavonoids, and steroidal saponins; and Cistanche's effects on male health are considered to be due to phenylethanoid glycosides, iridoids, and lignans. However, inconsistent with such traditional understanding, processing of these materials (e.g., processing ≤5‑year ginseng into red ginseng, processing Polygonatum into "Xianren Yuliang", and traditional drying of Cistanche) is often accompanied by a certain degree of polysaccharide degradation, partial conversion of saponins into aglycones, and loss of heat‑sensitive flavonoids. If these components were truly the functional basis, the efficacy of processed materials should be weakened due to decreased content of these active compounds. Therefore, there is reason to believe that during processing, components originally present in the raw materials with relatively weak activity may react with each other to form new components with strong physiological activity.

[0004] Based on the above, systematically analyzing traditional processing techniques/experience for food‑and‑medicine materials, confirming newly formed components and their mechanisms of action, and improving their content through precise control of processing conditions to strengthen health effects is of great significance for improving scientific processing, standardizing production, realizing controllable product quality, and protecting consumer health rights.

Summary of the Invention

[0005] Technical Problem
[0006] At present, traditional processing of food‑and‑medicine raw materials lacks systematic analysis, and in‑depth research is insufficient regarding compositional changes during processing and the functions of processed products. Therefore, it is necessary to explore the processing of traditional materials such as ginseng, Cistanche, and Polygonatum to understand compositional changes and optimize processing.

[0007] Technical Content
[0008] The present invention finds that ≤5‑year ginseng, Cistanche, and Polygonatum can all increase glucose and maltose contents through raw‑material pretreatment. Arginine is a dominant amino acid in all three materials after pretreatment, accounting for more than 50% of total free amino acids. In traditional processing, a heat‑processing stage typically occurs under relatively low system moisture (5%–25%) and relatively high temperature (95–110 °C). Such conditions favor the early‑stage Maillard reaction between free amino acids and reducing sugars, forming stable Amadori compounds (ACs) that may have physiological activity. During this stage, arginine in ≤5‑year ginseng, Polygonatum, and Cistanche reacts with maltose and glucose to form arginyl‑fructosyl‑glucose (AFG) and fructosyl‑arginine (FruArg). In particular, when the mass ratio AFG:FruArg is 5:1–1:3 and the sum of the two accounts for 2.5%–8% of product mass, the product shows a significant effect in improving aging‑related erectile dysfunction.

[0009] A first objective of the present invention is to provide a composition for alleviating male sexual dysfunction caused by aging, wherein the composition consists of arginyl‑fructosyl‑glucose (AFG) and fructosyl‑arginine (FruArg).

[0010] Further, the mass ratio of AFG to FruArg in the composition is 8:1–1:3.
[0011] Further, the mass ratio of AFG to FruArg in the composition is 6:1–1:3.
[0012] Preferably, the mass ratio of AFG to FruArg in the composition is 5:1–1:3.

[0013] Further, AFG may be prepared as follows: dissolve maltose and arginine in water, adjust solution pH to neutral, heat to react, and then purify and isolate AFG.

[0014] Further, the molar ratio of maltose to arginine is 0.5–1.5:1.
[0015] Further, the solution pH is 6.5–7.5.
[0016] Further, heating reaction is performed at 80–90 °C for 0.5–2 hours.
[0017] Further, purification may be column purification or gas chromatography purification; when using column purification, the resin may be Dowex 50WX4 ion‑exchange resin.

[0018] Further, FruArg may be prepared as follows: dissolve glucose and arginine in water, adjust solution pH to neutral, heat to react, and then purify and isolate FruArg.

[0019] Further, the molar ratio of glucose to arginine is 0.5–1.5:1.
[0020] Further, the solution pH is 6.5–7.5.
[0021] Further, heating reaction is performed at 80–90 °C for 0.5–2 hours.
[0022] Further, purification may be column purification or gas chromatography purification; when using column purification, the resin may be Dowex 50WX4 ion‑exchange resin.

[0023] A second objective of the present invention is to provide an application of the above composition in the preparation of medicaments.

[0024] A third objective of the present invention is to provide a processed product for alleviating male sexual dysfunction caused by aging, wherein the mass ratio AFG:FruArg in the processed product is 8:1–1:3.
[0025] Further, the mass ratio AFG:FruArg is 6:1–1:3.
[0026] Preferably, the mass ratio AFG:FruArg is 5:1–1:3.
[0027] Further, the processed product includes one or more of red ginseng, Cistanche, and Polygonatum.

[0028] A fourth objective of the present invention is to provide a method for preparing a processed product with high AFG and FruArg content and having a function of alleviating male sexual dysfunction caused by aging, comprising:
[0029] (1) adjusting moisture to 60%–80% and sealing;
[0030] (2) sealed incubation;
[0031] (3) drying to reduce moisture to 15%–40%;
[0032] (4) sealing and incubation;
[0033] (5) drying to below 10% to obtain the processed product.

[0034] Further, the raw materials are one or more of ginseng, Cistanche, and Polygonatum.
[0035] Further, the ginseng, Cistanche, and Polygonatum are cultivated for five years or less.
[0036] Further, raw materials may be whole, cut, pulverized, or mashed.
[0037] Further, when ginseng and Polygonatum are mixed, the ratio is 0.5–1.5:1.
[0038] Further, when ginseng and Cistanche are mixed, the ratio is 1.5–2.5:1.
[0039] Further, when Polygonatum and Cistanche are mixed, the ratio is 0.5–1.5:2.
[0040] Further, when ginseng/Polygonatum/Cistanche are mixed, the ratio is 0.5–1.5:0.5–1.5:1.
[0041] Further, moisture adjustment in step (1) comprises removing water at 50–60 °C.
[0042] Further, moisture is adjusted to 65%–80%.
[0043] Further, sealed incubation in step (2) is 50–60 °C for 48–72 h.
[0044] Further, drying in step (3) includes high‑temperature drying, natural air‑drying, or vacuum drying.
[0045] Further, high‑temperature drying is 90–105 °C.
[0046] Further, moisture is reduced to 15%–30% in step (3).
[0047] Further, the material in step (4) can be treated as described in claim 6.
[0048] Further, sealed incubation in step (4) is 90–110 °C for 30–150 min.
[0049] Further, drying in step (5) includes high‑temperature drying, natural air‑drying, or vacuum drying.
[0050] Further, high‑temperature drying is 90–105 °C.
[0051] Further, moisture after drying is 8%–10%.

[0052] Beneficial Effects:
[0053] By pretreating ginseng, Cistanche, and Polygonatum to increase glucose and maltose, and blending based on measured contents to obtain a combined material in which the dry‑basis glucose and maltose contents are each not less than 2%, their total dry‑basis content reaches 5% or more, arginine content is not less than 1.5% (dry basis) and arginine accounts for not less than 50% of total free amino acids, a material is obtained that can be processed through moisture control and sealed incubation into an ingredient in which the sum of AFG and FruArg is 2.5%–8% and the mass ratio AFG:FruArg is 5:1–1:3; experimental verification shows obvious effects in improving aging‑related erectile dysfunction.

Example 8 - Polygonatum + Cistanche Product
[0160] Mix Polygonatum:Cistanche = 1:2, mill; moisture 80%; incubate 50 °C 72 h; glucose 3.28%→4.88%, maltose 1.09%→2.49%, arginine 2.06%→2.35%; dry to 20% at 105 °C; seal incubate 105 °C 60 min; dry 105 °C to <10%; package.
[0166] Testing: AFG+FruArg 26.37; AFG 6.60; FruArg 19.77; ratio 0.33:1; 5‑HMF 0.0358; AA 0.0212 mg/g DW.

Comparative Example 1
[0168] Similar to Example 2 but without step (3) moisture reduction stage; yields AFG+FruArg 41.25; AFG 36.09; FruArg 5.16; ratio 6.99:1; 5‑HMF 0.0306; AA 0.0183.

Comparative Example 2
[0176] Same as Example 2 but in step (3) moisture adjusted to 50%; AFG+FruArg 57.23; ratio 6.08:1; 5‑HMF 0.0429; AA 0.0215.

Comparative Example 3
[0179] Same as Example 2 but in step (3) moisture adjusted to 10%; AFG+FruArg 30.78; ratio 8.41:1; 5‑HMF 0.0136; AA 0.0079.

Comparative Example 4
[0182] Same as Example 2 but in step (1) moisture adjusted to 90%; AFG+FruArg 69.31; ratio 5.45:1; 5‑HMF 0.0482; AA 0.0253.

Comparative Example 5
[0185] Same as Example 2 but in step (1) moisture adjusted to 50%; AFG+FruArg 61.81; ratio 5.93:1; 5‑HMF 0.0385; AA 0.0183.

[0187] Comparing Comparative Example 1 with Example 2 indicates that lacking step (3) results in decreased AFG+FruArg and an AFG:FruArg ratio that does not meet the needs described. Excessively high (Comparative 2) or low (Comparative 3) moisture in step (3) also reduces AFG+FruArg and leads to an unsuitable ratio.
[0188] Comparing Comparative 4/5 with Example 2 indicates moisture control in step (1) is also related to AFG+FruArg sum and the effect; too high or too low moisture reduces AFG+FruArg and affects the ratio.
[0189] Therefore, moisture control in steps (1) and (3) is critical to increasing AFG+FruArg sum and meeting AFG:FruArg between 5:1 and 1:3; step (1) should be 65%–80% and step (3) should be 15%–30%.

Example 9 - Efficacy Evaluation
[0191] Animals: 40 healthy male 8‑week‑old SPF SD rats.
[0192] Groups: C blank; D D‑gal model; P tadalafil 0.52 mg/kg; Sample‑L low dose (AFG:FruArg=5:1, 40 mg/kg sum); Sample‑H high dose (AFG:FruArg=1:3, 80 mg/kg sum).
[0193] D‑gal injection 150 mg/kg BW daily for 9 weeks; gavage from week 6 for 4 weeks.
[0194] Sample‑L from Example 2 red ginseng product; Sample‑H from Example 8 Polygonatum‑Cistanche product.
[0196] Results: no significant differences in body/penile weight; p53/p21 decreased significantly; erection frequency increased significantly and both sample groups outperformed positive control.
[0211] Conclusion: red ginseng product and Polygonatum‑Cistanche product rich in AFG and FruArg and within ratio 5:1–1:3 can alleviate male sexual dysfunction caused by aging.

[0212] The embodiments do not limit the scope; obvious improvements fall within the protection scope of the claims.

Title

Beyond Traditional Herbs: How Controlled Processing May Unlock New Bioactive Compounds in Cistanche for Men's Vitality Support

Who this is for

If you're a product developer, brand owner, or formulator exploring Cistanche (Cistanche deserticola / Cistanche tubulosa) for men's vitality, aging‑related performance support, or "healthy circulation" positioning, this is the processing insight you don't want to miss.

The core idea: processing can create new actives

Most herbal R&D focuses on "what's already in the plant": phenylethanoid glycosides in Cistanche, ginsenosides in ginseng, polysaccharides in Polygonatum, etc.

But the technical document you shared proposes a different hypothesis:

During traditional heat‑processing (steaming/drying), some "classic actives" may decrease-yet the final processed materials are still perceived as more functional. That mismatch suggests new compounds may be formed during processing and could be driving part of the activity.

The chemistry behind it: early‑stage Maillard reaction (the "useful part")

When you have:

Reducing sugars (glucose, maltose)

Free amino acids (especially arginine)

Heat and controlled moisture

…you can promote early‑stage Maillard reactions that form relatively stable Amadori compounds.

In this process, arginine can react with sugars to produce:

AFG = Arginyl‑Fructosyl‑Glucose

FruArg = Fructosyl‑Arginine

The document claims that a processed ingredient performs best when:

AFG + FruArg total: 2.5%–8% of product mass

AFG:FruArg mass ratio: 5:1 to 1:3

Why a Cistanche factory should care (practical product value)

From a manufacturing perspective, this isn't just "interesting chemistry"-it's a path toward:

Standardization beyond botanical variability (season, origin, polysaccharide drift)

A measurable marker system (AFG + FruArg content + ratio)

A processing method that can be described in SOPs (moisture targets, sealed incubation, temperatures, time)

This gives you something developers always ask for:

"What do we standardize to?"

"How do we keep batch‑to‑batch performance consistent?"

"What's your differentiator beyond 'high echinacoside'?"

The manufacturing "sweet spot": moisture is the control knob

The proposed process is essentially two controlled stages:

Stage 1 - Pretreatment to boost sugars (and keep arginine abundant)

Adjust moisture to 65%–80%, seal

Incubate 50–60 °C for 48–72 h

Purpose: increase glucose and maltose, while arginine remains a dominant free amino acid (the document notes arginine can exceed 50% of total free amino acids after pretreatment).

Stage 2 - Create AFG/FruArg under low‑moisture + heat

Dry to 15%–30% moisture

Seal and incubate 90–110 °C for 30–150 min

Final dry to <10% moisture (often 8%–10%)

Purpose: under these conditions, AFG and FruArg formation is favored.

Key takeaway for factories:
If you miss the moisture window (too wet or too dry), yields and ratios can drift. The comparative examples in the text show exactly that: moisture deviations reduce AFG+FruArg or push the ratio outside the proposed "optimal" range.

What should be tested (if you want real "developer‑grade" data)

If you want to build a credible EU/US product story, focus on analytics and safety, not only traditional markers.

1) Quantify AFG + FruArg (and ratio)

The text describes UPLC‑MS/MS conditions used to measure AFG and FruArg. Even if you change instruments, what matters commercially is:

validated method

reference standards

batch CoAs that report:

AFG (mg/g)

FruArg (mg/g)

sum and ratio

Sample-2 5.6 ± 0.77 c
Sample-3 5.85 ± 0.78 c
Sample-4 3.4 ± 0.47 b

2) Monitor heat‑process byproducts

The document mentions 5‑HMF and acrylamide (AA) tests in processed products. For Western markets, that's a strong trust signal: you are aware of thermal processing risks and you measure them.

3) Align with your intended regulatory lane

If you sell as dietary supplement / functional food ingredient, keep claims to "supports sexual health / vitality / healthy blood flow / performance under stress" depending on jurisdiction and substantiation.

Avoid "treats erectile dysfunction" or "drug‑like comparisons" on consumer pages.

Product development angles that work in the West (without risky claims)

Here are safer positioning frameworks:

Aging & vitality support: "supports male vitality as we age"

Performance confidence: "supports healthy performance and endurance"

Circulation support: "supports healthy blood flow" (be careful-still can be sensitive)

Stress + energy: pair Cistanche with adaptogen language where allowed

Suggested format for an ingredient spec sheet (what buyers want)

For B2B buyers, include:

Botanical ID + part used

Extraction vs. processed whole‑material (your differentiator)

Moisture specs (final 8%–10%)

Marker specs:

AFG + FruArg total % range

AFG:FruArg ratio range

Contaminant + safety panel (heavy metals, pesticides, micro, PAHs if relevant)

Thermal byproducts monitoring: 5‑HMF, acrylamide (if you choose to monitor routinely)
