Simultaneous Determination Of Echinacoside And 2,3,5,4'-tetrahydroxy Stilbene-2-Ο-β-D-glucoside in Congrong Degestive Oral Liquid By HPLC Ⅱ

2.4 Sample Content Determination

Ten batches of test samples were measured according to the method developed in this standard. The results are shown in Table 3. According to the above content measurement results, the content of echinaceaside in Cistanche Oral Liquid is 7.70~8.35 mg/mL, and the content of stilbene glycoside is 8.80~14.70 mg/mL. Cistanche tubulosa is a plant of the family Cistanche deserticola Y. C. Ma or Cistanche tubulosa.

The dry fleshy stems of tubulosa (Schenk) Wight with scaly leaves [6-8] are included in the 2020 edition of the Pharmacopoeia of the People's Republic of China [9]. The current pharmacopeia sets the content limits of echinacoside and verbascoside in Cistanche tubulosa as (1) The total amount of echinacea side (C35H46O20) and verbascoside (C29H36O15) in Cistanche tubulosa should not be less than 0.30%; 

(2) Cistanche tubulosa tubulosa

The total amount of echinaceaside (C35H46O20) and verbascoside (C29H36O15) shall not be less than 1.5%[10]. However, the content of verbascoside in Cistanche Oral Liquid is too low to be quantified, so only the content of echinaceaside is used as a quantitative index. The total theoretical amount of echinacoside and verbascoside is (750 g/1000 mL) × 0.3 %×1000=2.25 mg/mL, calculated based on Cistanche tubulosa tuberculate

The total theoretical amount of echinacoside and verbascoside is (750 g/1000 mL) × 1.5% × 1000 = 11.25 mg/mL. Determine the echinaceaside content of 8 batches of medicinal materials, produce 8 batches of samples accordingly, and measure the echinaceaside content. Thus, the average transfer rate of echinaceaside in the 8 batches of samples is calculated to be 23.7%, and the limit is the theoretical limit. The amount is 11.25 mg/mL.

ORGANIC CISTANCHE EXTRACT WITH 30% ECHINACOSIDE AND 12% ACTEOSIDE

Polygonum multiflorum is the dried root tuber of Polygonum multijiorum Thunb. Excavate the leaves when they are withered in autumn and winter, cut off both ends, wash them, cut them into large pieces, and dry them [11-12]. Calculated as a dry product, this product should contain no less than 1.0% stilbene glycoside (C20H22O9), and the theoretical amount is (1 500 g/1 000 mL) × 1.0% × 1000 = 15 mg/mL. The content of stilbene glycosides in 8 batches of medicinal materials was measured, and 8 batches of samples were produced accordingly. The content of stilbene glycosides was measured, and the average transfer rate of stilbene glycosides in the 8 batches of samples was calculated to be 36.8%, with a limit of The theoretical amount is 15 mg/mL × 37% = 5.6 mg/mL. Based on the content measurement results of the sample, it is preliminarily determined that this product contains polygonum multiflorum based on stilbene glycoside (C20H22O9), which should be no less than 6.0 mg/mL.

3 Discussion and conclusion

3.1 Investigation of extraction solvents

In this experiment, dilute ethanol, 70% ethanol, absolute ethanol, 50% methanol, 70% methanol, and methanol were used for extraction respectively to examine the effects of 7 extraction solvents on the content determination of Cistanche purpurea oral liquid. By comparing the contents of echinacoside and stilbene glycosides in 7 kinds of extraction solvent samples, it was found that each solvent has certain differences in the extraction rate of echinacoside and stilbene glycosides from Cistanche purpurea oral liquid. Dilute ethanol and 70% Methanol have little difference, and 70% methanol is slightly higher. Therefore, 70% methanol was selected as the extraction solvent for the determination of echinaceaside and stilbene glycoside content in Cistanche purpurea oral liquid [13-15].
