Abstract

The traditional Chinese medicine Cistanche cistanche series is a fleshy stem with scaled leaves of the family plant, which has the effect of nourishing the kidney and assisting yang. We used Hydrocortisone Acetate to make a mouse “Yang deficiency” animal model. Using the changes in the morphology and structure of the kidney and adrenal glands of the mice as indicators, we explored the protective effect of the alcohol extract of Roucong on the kidney and adrenal function.

Material

Hydrocortisone acetate injection: produced by Shandong Xinhua Pharmaceutical Factory, batch number: 921026.

Cistanche cistanche alcohol extract: Cistanche cistanche was purchased from a local medicinal material company, and the cistanche cistanche crystals were obtained by ethanol extraction.

Experimental animals: Experimental mice raised in the animal room of the 148th Hospital of the People's Liberation Army, both male, and female, weighing 22-28g.

Experimental method

There are 35 mice, 30 of which are made into Yang-deficiency animal models. Fifteen mice were treated with hydrocortisone acetate alone as the experimental group, 15 mice were treated with hydrocortisone acetate plus Cistanche extract as the drug group, and the other five were the normal control group.

Since the beginning of the experiment, the experimental group and the drug group have been injected with hydrocortisone acetate into the hip muscles at a dose of 0.7 mg/day per mouse. The drug group was given the alcohol extract of meat from the stomach via gastric tube at the same time, the dose was 15mg/day each. The three groups of animal experiment group, drug group, and normal control group were fed with the same volume of normal saline every day, and the experiment lasted 15 days. There were 7 deaths in the experimental group, 1 death in the drug group, and no death in the normal control group. A mouse was taken from the three groups and sacrificed. The kidneys and adrenal glands were immediately dissected. The tissues were fixed with formalin solution and routine pathological examinations were performed.

Results

In the experimental group, the adrenal cortex was atrophied, and the fascicular zone cells decreased in size. The tubular epithelial cells of the kidney are swollen, and casts can be seen in the distal convoluted tubules. In the drug group, there was no atrophy of the adrenal cortex, the thickness of the fascicular zone cells was close to normal, and there was no abnormality in the kidneys. Normal control group Adrenal cortex and kidney are normal.

The above experimental results show that when animals use corticosteroids for a longer period of time, the addition of an alcoholic extract of meat from the traditional Chinese medicine for nourishing the kidney and promoting yang can prevent the adrenal cortex atrophy caused by the single use of hormones. In addition, the alcohol extract of meat from puree also has a certain protective effect on kidney function.
