Abstract

Plant immunity is activated upon pathogen perception, and when it is constitutively active, it usually affects growth and yield. How plants fine-tune immune homeostasis in their natural habitat remains elusive. Here, we identified a conserved immunosuppressive network that coordinates immune homeostasis in cereals, centered on the Ca2+ sensor, and rice resistance to disease (ROD1).ROD1 promotes reactive oxygen species (ROS) scavenging by stimulating peroxidase activity, and its protein stability is regulated by ubiquitination. Disruption of ROD1 confers resistance to a wide range of pathogens, and rice with an agroecological-specific distribution of the natural ROD1 allele prevalent in the specific distribution of the natural ROD1 allele prevalent in Indica rice enhances resistance without affecting yield. The fungal effector AVRPI -t mimics ROD1 structurally and activates the same ROS clearance cascade to suppress host immunity and promote virulence. Thus, we reveal a molecular framework adopted by the host and pathogen that integrates Ca2+ sensing and ROS homeostasis to suppress plant immunity, which provides a principle for breeding disease-resistant, high-yielding crops.

Introduction

we conducted a long-term study to track rice susceptibility to genetic resistance to disease1 (ROD1).ROD1 encodes a C2 structural domain Ca2+ sensor that acts as a global regulator of immunity. We found that ROD1 promotes H2O2 degradation by activating the peroxidase CatB. the stability of ROD1 is regulated by a pair of E3 ubiquitin ligases, RIP1 and APIP6. loss of function of ROD1 results in robust broad-spectrum resistance to a variety of bacterial and fungal pathogens. rodd1 has a six-stranded antiparallel b-sheet structure that is also found in the fungal blast effector AVRPI - t. ROD1 and AVRPI -t share the same protein degradation and ROS elimination cascade, supporting a model of fungal effectors exploiting host protein-mediated immune regulation. We identified a naturally occurring, locally adapted ROD1 allele that confers higher basal resistance without loss of crop yield, highlighting the importance of this factor in balancing immunity and growth. Thus, our study reveals a previously unrecognized mechanism by which plant and pathogen factors suppress plant immune responses through the same cascade.
