2. 9. Inspection of the extraction process of medicinal materials. 

After collecting information and reviewing a large number of papers and journals, 3 factors including material-to-liquid ratio (A), solvent concentration (B), and extraction times (C) were selected as the factors to be investigated for both Grasshopper and Cistanche deserticola. The results of the range analysis show that the order of influencing factors of Grasshopper is: solvent concentration > extraction times > solid-liquid ratio. Therefore, the optimal extraction conditions of Cistanche deserticola total glycosides are determined as A2B3C1, that is, the optimal extraction plan is to extract once with 90% ethanol, and the solid-liquid ratio is 1:20; the order of influencing factors of Cistanche deserticola is: solid-liquid ratio > number of extractions > Solvent concentration.

Therefore, the optimal extraction conditions A2B3C2 were determined, so the optimal extraction plan was a solid-liquid ratio of 1:20 and extraction twice with 90% ethanol solution.

2. 10 Selection of wavelength for determination of total glycosides

After scanning the absorption wavelength of the control substance and the test substance at 400 to 800nm, the results show that there is little difference in the absorption value of the salidroside reference substance between 520nm and 580nm, while after the color development of Cachinensis and Cistanche deserticola, The wavelength scanning results show that both have maximum absorption near 523nm. After browsing a large amount of literature, considering the actual conditions and the comprehensive chemical components of Grasshopper and Cistanches, we finally selected salidroside as Grasshopper. and the reference substance for the determination of total glycosides of Cistanche deserticola, and the detection wavelength after color development was determined to be 523nm.

2. 11 Investigation on extraction methods of medicinal materials and extraction solvents

In the early stage of the experiment, different extraction methods such as heating reflux, Soxhlet extraction, ultrasound, and cold-soaking were compared. The results showed that the amount of total glycosides obtained by the heating reflux extraction method was higher than that of other methods, so the heating reflux method was chosen to extract grass. Considering the polarity and solubility of phenylpropanoid saponins, three extraction solvents of methanol, ethanol, and n-butanol were investigated and analyzed. The results showed that the extraction effect of ethanol was better, so ethanol of different concentrations was used as the extraction solvent in this experiment.


3 Discussion

In this experiment, salidroside was used as the reference substance for the first time to determine the total glycoside content in Grasshopper and Cistanche deserticola. The total glycoside content in Grasshopper sample was 10. 61%, the total glycoside content in Cistanche deserticola sample is 10. 07%. Although spectrophotometry is a classic and mature content determination method, the color reaction between total glycosides and vanillin-glacial acetic acid and perchloric acid is also very sensitive, but the detector requires accurate and fast operating techniques to ensure satisfactory results in the experiment. .

In summary, this method has high precision, simplicity and good stability, and can be used for the determination of total glycosides content in Grasshopper and Cistanche deserticola. The chemical composition and pharmacological effects of Cistanche deserticola and Cao Congrong are similar, which provides a theoretical basis for the historical supplementary use of Cistanche deserticola and Cao Congrong, suggesting that the development and utilization of Cao Congrong has broad application prospects. The extraction process and biological activity of the two total glycosides are being further studied.
