Abstract: Objective To observe the effect of Cistanche decoction on the anti-fatigue ability of exercise mice. Methods The effects of Cistanche decoction on the anti-fatigue ability of exercise mice were evaluated by comparing the residence time, total ATPase activity, and related biochemical indexes of experimental animals on the automatic fatigue rotating rod instrument. Results Cistanche decoction can increase the body mass of mice, prolong the exercise time of mice, increase hemoglobin content, reduce blood lactic acid content after exercise, and enhance total ATPase activity. Conclusion Cistanche decoction can improve exercise endurance and accelerate the elimination of fatigue.

Cistanche herb is a parasitic plant belonging to the genus Orobanchaceae Cistanche, which is mainly produced in my country's Inner Mongolia, Xinjiang, Gansu, and Ningxia. Because it grows in the desert and has excellent medicinal value, it is known as the "desert ginseng". Cistanche has the functions of nourishing essence and blood, nourishing the kidney and strengthening tendons, moisturizing the five internal organs, warming the waist and knees, and curing internal injuries due to fatigue. Li Shizhen said: "This thing is tonic but not serious, so it has the title of calmness. Smooth, gentle and calm, and gentle appearance." According to statistics, Cistanche is the first in the prescription of traditional Chinese medicine for increasing strength. The purpose of this study is to preliminarily explore the role of Cistanche decoction in anti-fatigue, and provide a theoretical and experimental basis for the development of Cistanche as an anti-sport fatigue drug or sports health drink.

1 Material and methods

1.1 Experimental animals and drugs

30 SPF male Kunming mice, weighing 18-20g, were purchased from the Laboratory Animal Center of Lanzhou University Medical Department, the production license number is SCXK (甘)20050007, and the animal qualification number is 0000285. The decoction of Cistanche is homemade, with a mass concentration of 1.0 g/ml.

1.2 Experimental equipment

Electronic scale (JY4001, Shanghai Balance Instrument Factory), fatigue rotating rod instrument (ZB-200, Chengdu Taming Technology Co., Ltd.), automatic biochemical detector (SYNCHRON LX20, Chongqing Take Huayi Pharmaceutical Co., Ltd.), total ATP The enzyme detection kit was purchased from Nanjing Jiancheng Institute of Biological Engineering.

1.3 Experimental method

After 30 mice were adaptively fed for 3 days, they were randomly divided into 3 groups, each with 10 mice, namely the control group (normal saline), the Cistanche high-dose group (1.0g/mL), and the low-dose meat puree group ( 0.25 g/mL). Gavage was given continuously for 30 days, twice a day, 0.02 rnL/g each time, the control group was given the same amount of normal saline, and related indicators were detected after 30 days.

1.3.1 Body mass measurement

After 30 days, the body weight was weighed with an electronic scale and compared with the body weight before the administration.

1.3.2 Anti-fatigue test

First adjust the fatigue rotating rod instrument to the training state, put the mice on the adaptive training for 10 minutes, then adjust the fatigue rotating rod instrument in the test state, the speed is 30 r/min, and put the trained mice on the rotating rod in turn On the top, record the time that the mouse stays on the fatigue rotating rod instrument. 1.3.3 Biochemical index detection

After exercise, the mice immediately took their eyeballs and took blood. The automatic biochemical analyzer detected the hemoglobin (Hb) content and the blood lactic acid (LD) content, and the kit detected the serum total ATPase activity.

1.4 Data processing

All data were processed with SPSS13.0 software, expressed as £±S, and comparisons between groups were used and tested.

2 Results

2.1 The effect of Cistanche decoction on the body mass of mice

It can be seen from Table 1 that the bodyweight of the mice increased significantly after 30 days of the administration, and the differences between the treatment groups and the control group were statistically significant (P <0.05 or P <0.01), and the bodyweight of the high-dose group increased more significantly.

2.2 The effect of Cistanche decoction on the anti-fatigue ability of mice

The effect of Cistanche decoction on the anti-fatigue ability of mice was measured by a fatigue rotating rod instrument, and it was found that the anti-fatigue ability of the mice in the administration group was significantly enhanced compared with the anti-fatigue ability of the mice in the control group (P <0.05 or P <0.01), the effect of Cistanche high-dose group and the lower-dose group is more obvious; Cistanche high-dose group and the low-dose group have a dose-dependent fatigue effect than that of the low-dose group. The results are shown in Table 2. It is statistically significant (P <0.05), indicating that the anti-fatigue ability of Cistanche decoction is effective.

2.3 The effect of Cistanche decoction on the biochemical indicators of exercise mice

It was found through testing that the blood hemoglobin content of the mice in the administration group was significantly increased compared with the control group, the content of lactic acid was significantly reduced, and the activity of ATPase was significantly enhanced (P <0. 05 or F<0.01), indicating that the blood's ability to transport oxygen is enhanced, and energy metabolism is improved. Among them, the effect of high-dose Cistanche decoction is more obvious. The results are shown in Table 3.

3 Discussion

The traditional Chinese medicine Cistanche was first published in "Shen Nong's Materia Medica" and was listed as top grade. The cursive scripts of the past dynasties, such as "Wu Pu's Materia Medica", "Materia Medica Annotations", "Pictures of Materia Medica", "Rihuazi Materia Medica" and "Famous Doctors" etc. have recorded them. Cistanche is indispensable in the many tonic prescriptions and health medicinal wines in ancient my country. For example, "cistanche tubolosa extract", "cistanche supplement", and "cistanche herb and Cistanche decoction" named after Cistanche have dozens of different recipes; while Cistanche is cooked with rice, "female rice," and "female rice is stewed with meat." "Meat" is a good folk tonic.

At present, the main endurance test used for the evaluation of exercise fatigue is the endurance test. Measuring the time from continuous exercise to exhaustion can reflect the endurance of the body. It has been reported in the literature that Cistanche can extend the swimming time of mice, reduce the increase in serum creatine kinase after load exercise, and keep the ultrastructure of mouse skeletal muscle normal after exercise, indicating that Cistanche has the effect of enhancing physical strength and anti-fatigue [3F; Japan's Sato. T et al. (5) Experiments confirmed that Cistanche has the ability to counteract the weakening of autonomous function activities caused by the suspension stress load. In this experiment, the residence time of the mouse on the fatigue rotating rod instrument was used as an index to judge the endurance of the mouse. It was found that the Cistanche decoction can increase the anti-fatigue ability of the mouse, and the effect is dose-dependent, which is consistent with the literature report. . The direct source of energy required by the human body during exercise is the decomposition of adenosine triphosphate (ATP). The synthesis of ATP can be carried out in two forms of energy supply, namely anaerobic metabolism and aerobic metabolism, under these two energy supply forms There are three energy supply systems working, namely high-energy phosphate system (ATP-CP system), lactic acid system (anaerobic glycolysis system) and aerobic oxidation system. Anaerobic metabolism means that when the human body exercises in a state of insufficient oxygen supply, the energy supply is provided by the anaerobic decomposition of substances, which is mainly supplied by the ATP-CP system and anaerobic glycolysis system, which produces less energy and metabolites. When lactic acid accumulates to a certain amount, the system can be inhibited by feedback. When the human body exercises under the condition of sufficient oxygen supply, the energy supply is supplied by the thorough oxidation of energy substances. At this time, the working energy supply system is an aerobic oxidation system, which can generate a large amount of energy.
