Cistanche is a traditional and precious Chinese medicine. Its medicinal part is its dry fleshy stem with scales and leaves. Cistanche is mainly produced in Xinjiang, distributed in both northern and southern Xinjiang, and is the main producing area of Cistanche. Total Glycosides of Cistanche is a class of Phenylethanol Glycosides compounds extracted and isolated from plants in the genus Cistanche. It is the main effective part of plants in the genus Cistanche. At present, nearly 70 monomer components have been isolated from plants in the genus (Liu Guojun, editor in chief. Cistanche And artificial planting. China Labor and Social Security Press, 2003.120). There are hundreds of research literature on Total Glycosides of Cistanche at home and abroad, but none of them involve the effect of inhibiting skin pigmentation and whitening beauty [Deng Yan et al. Research progress on the regulation of melanocytes and tyrosinase by Chinese medicine. New Chinese Medicine, 2003,35(5):72-73]o The present invention takes Glycoside from cistanche tubulosa as the research object and uses the tyrosinase dopa rate oxidation method to study its tyrosinase inhibitory effect. The results show that Total Glycosides of Cistanche have a significant down-regulation effect on tyrosinase activity, which suggests that total Glycosides of Cistanche have the effect of inhibiting skin pigmentation and whitening skin effects.

Cistanche inhibits skin pigmentation

Tyrosinase is the main rate-limiting enzyme in the process of skin melanin biosynthesis. It is a complex of copper and protein. It can hydroxylate tyrosine, the main raw material for melanin production in the body, to produce L-dopa, which is then oxidized to dopa dai. Dopa dai undergoes a series of metabolic processes and finally combines with protein to produce melanin. The most important factor of skin color, excessive synthesis can cause the formation of hyperpigmented skin diseases such as freckles, chloasma, age spots, and melanoma. Therefore, through studies on the inhibition of tyrosinase activity, effective ingredients that inhibit skin pigmentation can be screened out, which can be applied to the prevention of hyperpigmented skin diseases and the skin whitening and freckle removal function of external cosmetics.

Summary of the invention:

The purpose of the present invention is to provide a new use of Total Glycosides of Cistanche or an extract containing Phenylethanol Glycosides of Cistanche. The method used is to observe the inhibitory effect of Total Glycosides of Cistanche on tyrosinase by using the tyrosinase rate oxidation method. The result is that Total Glycosides of Cistanche have a significant down-regulation effect on tyrosinase activity, suggesting that it can inhibit skin pigmentation And whitening and beauty functions, providing new functional positioning for Cistanche health care products and pharmaceutical preparations, and expanding its application scope to the field of cosmetics.

Test method:

The specific method for detecting the specific absorbance value A490o of the reaction product at 490nm using the tyrosinase dopa rate oxidation method with an enzyme-linked immunoassay is as follows:

1. Mix tyrosine (Tianjin Guangfu Fine Chemical Research Institute) into a 0.02% solution with 67mmol • L'* 12 phosphate buffer (PB solution), and tyrosinase (2350U/mg, Sigma company) into a solution 500U-mL'1 solution, bear fruit shake (Beijing Naike Biochemical Technology Research Institute) is made into 1.0mg•mL4 solution.

2. Total Glycosides of Cistanche come from Cistanche (Cista&che tubidosaR.Wizheng), measured by ultraviolet spectrophotometry, and contains more than 50% of phenethylamines. Prepare Total Glycosides of Cistanche with 67mmol • L'1 phosphate buffer solution to a concentration of 0.5~3.0mg • mL'1.

3. The samples without drugs were used as the blank control group, and Xiong Guoyu was used as the positive control group.

4. The reaction is carried out in a 96-well culture plate. The total reaction system is 150lxL. Take 50 nL of the test substance and add 50 uL of tyrosine. After fully mixing, quickly add 50uL (25U) of tyrosinase in an ice bath and mix. Evenly, incubate at 25°C for 15min, and immediately set the microplate reader to measure A490 in each well. Each group of parallel tests 5 times.

Test results:

Different concentrations of Total Glycosides of Cistanche have different degrees of inhibition on tyrosinase, and the inhibition rate is roughly dose-dependent with the dose of the effector within a certain concentration range, which is expressed under the same substrate concentration condition, The inhibitory effect of Total Glycosides of Cistanche on tyrosinase increases with the increase of concentration. The concentration of Total Glycosides of Cistanche is in the range of 0.5~3.0mg-mL-1, which has an inhibitory effect on tyrosinase activity, and there is a good dose-effect relationship in the concentration range of 0.5~2.0mg • mL; Total Glycosides of Cistanche and Under the same concentration of bearberry, the effect is not as good as that of bearberry, but when the concentration is increased to 1.5mg·mL, the effect is stronger than that of bearberry. After the mean t-test of the two samples, the Total Glycosides of the Cistanche group (concentration in the range of 1.0~3.0mg-mL'1) and the positive control bearberry alfalfa group were compared with the blank group, and the inhibitory effect on tyrosinase was statistically significant. The scientific significance, that is, Total Glycosides of Cistanche and bearberry smoothes have a significant down-regulation effect on tyrosinase activity, suggesting that Total Glycosides of Cistanche has the effect of inhibiting skin pigmentation and skin whitening effects, and it will be developed as a treatment for hyperpigmentation in the future. Dermatological drugs and whitening and freckles removing ingredients as external cosmetics lay a good foundation.
