The effect of Cistanche on urination of healthy rats and rats with kidney failure

Contrary to the effect on Healthy rats, giving rats continuous ig Cistanche for 10 days significantly reduced the increase in urine output within 0～1 hour and 1～2 hours after the last dose of Kidney failure rats, and increased the urination of Kidney failure rats. Bodyweight and some organ weights. Conclusion: Cistanche has different regulating effects on the urination of Healthy rats and Kidney failure rats.

Cistanche Y.C.Ma is the fleshy stem of Cistanche, traditional Chinese medicine for improving kidney function. It has the effects of nourishing the kidney, nourishing essence and moisturizing the intestines. "Materia Medica" is called the essential medicine for replenishing essence and blood and nourishing the kidney. It has a good effect on fatigue, forgetfulness, hearing loss, and enuresis caused by kidney deficiency. Cistanche is also used in folks to treat senile urinary disorder (frequent urination, urinary frequency, urination). Urgency, dripping urine, etc.). Modern studies have shown that Cistanche has certain anti-aging effects [1, 2], and its total extract has the effects of enhancing the body's immune function [3, 4] and inhibiting lipid peroxidation [4, 5]. There have been reports of experimental studies on the effects of Cistanche [91], but no reports of its effects on animal urination have been seen. This article discusses its effect on urination in Kidney failure rats.
1 material
1.1 Test drugs:
Cistanche pastes 2.5 g/ml, batch number 901127, provided by the Natural Medicine Laboratory of China Medical Research and Development Center.
1.2 Animals
Do not use Wistar rats, purchased from the Animal Department of Beijing Medical University.
2 Methods and results
2∙1 Effect on the urinary output of Healthy rats
In order to reduce the individual differences in the urination of animals, “rats ig normal water 2∙5ml/100g and then collect the volume within 2 hours. ‚Choose those whose urination volume reaches more than 40% of the gavage volume as experimental animals. Thirty male rats weighing 231±19g were divided into 3 groups according to their body weight. ‚After 24 hours of fasting and water, ig 3.0ml/100g of normal saline and squeeze the rat’s lower abdomen after loading for 30 minutes to empty the bladder as much as possible. Then, the animals in each group were given a liquid medicine or an equal volume of normal saline at a bodyweight of 2.0ml/100g. Place the animals in metabolic cages, one for each cage, and collect the urine of 0～1, 1～2, 2～4, 4～6 hours after administration respectively, and measure the volume. The results showed that the urinary volume of the Cistanche group was significantly higher than that of the control group within 2 hours (0～1h, 1～2h) after administration (see Table 1).

2∙2 Effect on urination of rats with kidney failure
40 male rats weighing 350±40g, divided into 4 groups according to body weight, except for the blank control group, im sterilized saline 0.3ml/100g per day, all other groups were injected with an equal volume of 0.5% hydrocortisone At the same time, administer medicine or an equal volume of normal saline 1∙0ml/100g for 10 days. 30 minutes before the last dose, ig 3.0ml/100g normal saline load for 30 minutes, squeeze the rat's lower abdomen to empty the bladder as much as possible, and then administer. ‚After the drug, put a mouse in each cage and place the animal After being placed in the metabolic cage, the urine output was collected for 0 to 1 hour, 1 to 2 hours, 2 to 4 hours, and 4 to 6 hours after administration. Then weigh the body weight, and weigh the thymus, adrenal glands, prostate, seminal vesicles, testes, and bladder after slaughter. The results showed that the urinary output of Cistanche at 0～1 hour and 1～2 hours was significantly lower than that of the hydrocortisone group; the urination output of Cistanche 10g/kg group at 2～4 hours and 4～6 hours was lower than that of hydrocortisone (see Table 2). The weight of rats in the hydrocortisone kidney failure group and the weight of thymus, adrenal glands, testes, and other organs were significantly reduced. Cistanche can significantly increase the weight, adrenal glands, seminal vesicles, and testes of the kidney failure rat weight (see Table 3).

3 Discussion
Traditional Chinese medicine believes that “the kidney governs water,” the gasification of the bladder, the triple-burner and the excretion of water and liquid all depend on the temperature of the kidney. Loss of urinary control (frequent urination, urgency, incessant urination, nocturia, etc.) is caused by insufficient kidney yang and cold in the lower Jiao. It is more common in middle-aged and elderly patients. ‚Therefore, the treatment of out-of-control urinary should focus on improving kidney function. After intramuscular injection of hydrocortisone in rats, the bodyweight of the kidney failure model group was significantly reduced, and the adrenal and gonadal coefficients were significantly reduced. ``And the converted urine output in the 0~1 hours (the animal weight difference between each group is significant, so it needs to be converted by weight) compared with the Healthy control group of rats. ig Cistanche has an obvious anti-weight loss, adrenal and gonadal coefficient decline and other symptoms of kidney failure. ‚The urinary output of the two-dose groups of Cistanche within 0 to 1 hour and 1 to 2 hours after administration was significantly lower than that of the kidney failure group, and there was a certain dose negative correlation. The urinary output of the Cistanche 10g/kg group in 2～4h and 4～6 hours was higher than that of the hydrocortisone group, which may be caused by the weakening of the drug effect.
The results of the study with Healthy rats show that "Cistanche has the effect of promoting urination in Healthy rats. "This is consistent with our previous report [7] that Cistanche promotes bladder urine emptying. ‚Contrary to the result of urination on kidney failure rats. Cistanche has two completely different effects on the urination of Healthy rats and kidney failure rats. ``First, it should be explained from the increase in urine output of rats after kidney failure: rat im hydrocortisone can cause kidney failure. Rats have increased urine output, which may be consistent with some symptoms of clinical urinary out-of-control. Cistanche causes kidney failure Animals have reduced urine output, "This is consistent with traditional Chinese medicine that Cistanche improves kidney function. Of course, whether the increase in urine output after the kidney failure caused by hydrocortisone is related to clinical urinary out-of-control symptoms still needs to be confirmed by further experiments. "If the result is positive," this may also be a good animal pharmacological model for urinary out-of-control diseases in traditional Chinese medicine.
Secondly, it has a promoting effect on Healthy animals. Although the animals used are awake rats and the anesthesia rats used in previous reports [7] are in a different state, "These two tests are whole animals, acute short-term tests, so they are effective The mechanism should be consistent, and the same result is expected. Cistanche has two opposite effects on different animals (one is Healthy and the other is the sick animal model). “This may also be proof that traditional Chinese medicine pays attention to syndrome differentiation and treatment.
