Abstract
Aging is the primary risk factor for many neurodegenerative diseases. Kidney-tonifying Chinese herbs, including Cistanche, Cornus Officinalis, and Epimedium, have been reported to show beneficial effects on several aging-related diseases. Based on recent studies, the neuroprotective and anti-inflammatory effects might explain the possible mechanisms of kidney-tonifying Chinese herbs on aging and central nervous system disorders.

Kidney-tonifying Chinese herb Cistanche
Over the past century, the average human life expectancy has increased especially in developed countries. As aging impairs cognitive, motor, and sensory functions, longer human lives have led to a global burden of aging-related diseases[1. Aging is the primary risk factor for many neurodegenerative diseases. including Alzheimer's disease (AD).Parkinson's disease (PD) and another central nervous system (CNS) disorders[2]. For most neurodegenerative diseases, there are few or no effective treatments, which tend to progress into more severe conditions and induce the disabilities of the elderly. Recently. kidney-modifying Chinese herbs have been reported to show beneficial effects on several aging-related diseases.
According to the theory of traditional Chinese medicine (TCM), the "kidney" shows a close connection with the function of the CNS. Many kidney-modifying Chinese herbs were found to modify the aging condition and neurological disorders. Cistanche is a Yin-nourishing and kidney-tonifying prescription in traditional Chinese medicine with a long history. Cistanche shows therapeutic effects on aging-related diseases, such as cancer, cognitive dysfunction, depression, and cardiovascular disease, by modifying the neuroendocrine immunomodulation network[3].
Cormus officinalis Sieb.et Zucc.(Shan-Zhu-Yu),one of the herbs in Cistanche is used to tonify the liver and the kidney for the treatment of vertigo, aching back, and sweating. Cornel iridoid glycoside is the active ingredient extracted from Cormus officinalis. In our previous studies, we found that cornel iridoid glycoside also increased the life span and senescence status in senescence-accelerated mouse-prone 8 mice[4]. Moreover, cornel iridoid glycoside inhibited inflammation and apoptosis, promoted neurogenesis and angiogenesis, and improved neurological function in the animal models of brain iniury[5-81. In animal models of AD.cornel iridoid glycoside improved the cognitive impairment, neuronal and synaptic dysfunctions via inhibiting tau pathology[9-11]. These results show the possible use of Cormus officinalis and cornel iridoid glycoside for retarding aging and treating neurological disorders.
Epimedium (Yin-Yang-Huo) is the dried leaf of Epimedium brevicormu MaximEpimedium sagittatum(Sieb. & Zucc.)Maxim,Epimedium pubescens Maxim.,or Epimedium ko-reanum Nakai. Epimedium is commonly used in traditional Chinese medicine as a kidney-tonifying herb. Epimedium flavonoids, the main component extracted from Epimedium, have been reported to delay senescence and show neuroprotective effects on aging and CNS disorders[12-15]. Our recent studies found that epimedium flavonoids showed beneficial effects on a rat model of cerebral small vascular disease by protecting both the white matter and the grey matter in the brain[l6.17], Icariin is the main active component in epimedium flavonoids. We found that icariin enhanced the remyelination process in a cuprizone-induced demyelination mouse model by promoting oligodendrogenesis and neurotrophic factor production[ 18]. Moreover, both epimedium flavonoids and icariin promoted the proliferation and differentiation of neural stem cells in vitro[19,20]. These results indicate the possible use of Epimedium for delaying aging and treating CNS disorders.
Based on the theory of traditional Chinese medicine and the results of modern pharmacology, we found the anti-aging properties of kidney-tonifying Chinese herbs, especially in CNS disorders. Shen-Jing (Kidney essence)is considered as the origin of life. During aging or injuries, Shen-Jing might be destroyed or show deficiency, Shen-Jing deficiency might cause the dysregulation of neuroendocrine immunomodulation network, causing inflammation and deficiency in regeneration (stem cells)[21]. Thus, our group explained the possible mechanisms of kidney-tonifying Chinese herbs on aging and CNS disorders, including promoting energy metabolism, nourishing the neuronal cells, increasing the expression of neurotrophic factors, and regulating inflammation[22]. Overall, the neuroprotective and anti-inflammatory effects might explain the possible mechanisms of kidney-tonifying Chinese herbs on aging and CNS disorders. More studies of this kind of herbs on aging are needed to meet the global burden of aging-related disorders.
