Abstract

Cistanche series is a fleshy stem with scaled leaves of the family plant, which has the effect of nourishing the kidney and aphrodisiac. We used hydrocortisone acetate to make a mouse "Yang deficiency" animal model [1], taking the morphological and structural changes of the kidney and adrenal gland of the mouse as indicators to observe the protective effect of the ethanol extract of Cistanche on the function of the kidney and adrenal gland, summarized as follows.

Experimental method

Thirty-five mice were made into yang-deficiency animal models (15 mice were treated with hydrocortisone acetate alone as the test group, and 15 mice were treated with hydrocortisone acetate plus the alcohol extract of Cistanche as the drug group), and the other 5 mice It is a normal control group. The experimental group and the drug group were injected intramuscularly with hydrogen acetate test pine at a dose of 0.7 mg/d each. The drug group was given the alcohol extract of Cistanche via gastric tube at the same time, the dose was 15mg/d each. Three groups of animals (experimental group, drug group, and normal control group) were fed with the same volume of normal saline every day, and the test lasted for 15 days (7 deaths in the experimental group, 1 death in the drug group, and no death in the normal control group), 3 groups of animals Five mice were sacrificed, and the kidneys and adrenal glands were dissected immediately. The tissues were fixed with formalin solution and routine pathological examinations were performed.

In the experimental group, the adrenal cortex was atrophied, and the cell volume of the fascicular zone decreased. The tubular epithelial cells of the kidney swell and casts can be seen in the distal convoluted tubules. In the drug group, there was no atrophy of the adrenal cortex, the thickness of the fascicular zone cells was close to normal, and there was no abnormality in the kidneys. The adrenal cortex and kidney of the normal control group were normal. The test results show that when animals use corticosteroids for a longer period of time, adding alcohol extracts of Cistanche cistanche, a traditional Chinese medicine that nourishes the kidney and yang, can prevent the adrenal cortex atrophy caused by the single use of hormones. The alcohol extract of Cistanche has a certain protective effect on kidney function.
