Abstract

Purpose: To clarify whether Cistanche cistanche can be used as a substitute for Cistanche. Method: According to its main therapeutic effects, the decoction of Cistanche cistanche and Cistanche tubulosa decoction on mice's laxative effect and the weight and cold tolerance time of mice with kidney yang deficiency 0 were carried out. Result: Both of them have certain functions of laxative and moisturizing and warming and invigorating kidney-yang. Conclusion: It is preliminarily believed that Cistanche cistanche can be used instead of Cistanche when the resources of the authentic medicinal materials of Cistanche are rapidly decreasing.

Cistanche is a commonly used Chinese medicine for invigorating the kidney and yang and has the functions of invigorating the kidney, replenishing essence, and moisturizing dryness. 5 The 62000 edition of the Chinese Pharmacopoeia contains the dried fleshy stems with scales of Cistanche deserticola Y. C. Ma, a plant of the family Cistanche deserticola Y. C. Ma, as a genuine medicine. Due to the particularity of the parasitic growth and reproduction of Cistanche, combined with its remarkable curative effect, it has been artificially indiscriminately dug and its host Haloxylon ammodendron has been cut down in large numbers, which has caused the resources of Cistanche to decrease sharply, and it has become an endangered plant 112. Due to the shortage of medicine sources, other varieties of this genus are also used everywhere. Among them, Cistanche tubulosa (Schenk) R. Wight is mainly produced in Xinjiang, China, and the resources are relatively abundant. It is the main source plant of Cistanche tubulosa (Schenk) R. Wight at present.122. To clarify whether the Cistanche Tubulosa can be used as a substitute for Cistanche, we conducted a comparative study on the pharmacological effects of Cistanche and Cistanche decoction on mice with laxative and on/kidney-yang deficiency 0 mice based on its main therapeutic effects.

Experimental Materials

Medicinal materials

Cistanche and Cistanche tubulosa were purchased from Alexa Banner in Inner Mongolia and Hetian in Xinjiang and have been identified.

Reagent

1% phenol red aqueous solution and 0.8 mg/ml hydrocortisone solution were prepared with physiological saline.

Sample processing

Take 100g slices of Cistanche cistanche and Cistanche tubulosa respectively, grind into a coarse powder, and boil 3 times, respectively for 2 hours, 1.5 hours, and 1 hour, combine the three filtrates and concentrate them to the equivalent of each ml. 1g crude drug and each ml are equivalent to two concentrations of 0.5g crude drug.

Kunming mice, provided by the Experimental Animal Center of Guangzhou Medical College.

Methods and results

Comparison of defecation effects in mice. Add 1% phenol red to the medicine decoction. A total of 60 Kunming mice weighing 20? 2g were divided into 5 groups randomly: model, Cistanche cistanche, and Cistanche cistanche group. Fasting without food for 24 hours, the control group was given 1% phenol red solution, and the other 4 groups were given respectively with Cistanche cistanche 0.5g/ml and 1g/ml decoction, and Cistanche tubulosa 0.5g/ml, 1g/ml water. Decoction, 0.2g/10ml was administered to each animal and timed immediately after administration. Each group recorded the time from the end of the administration to the discharge of the first red feces for each mouse and observed the hardness, properties, etc. of the feces.


After taking the drug, the animals quickly excreted red urine. The animals in the administration group had a shorter time to excrete red feces, and the feces surface was shiny, soft, and shaped. In the 0.5g/ml dose group of Cistanche and Cistanche tubulosa, the defecation time was shorter than that of the control group, and there was a significant difference (P <0.05); the 1g/ml dose group, the defecation time was very significant compared with the control group The difference (P <0.01) shows that both Cistanche and Cistanche tubulosa have obvious laxative effects; and with the increase of the dose, the defecation time is also significantly different (P <0.05), but Cistanche and tube Compared with the defecation time of the same dose of Cistanche cistanche, there was no significant difference (P>0.05).


Comparison of cold tolerance in hydrocortisone/kidney-yang deficiency mice 132 60 male Kunming mice weighing 28-2g was selected and randomly divided into 6 groups with 10 mice in each group. In this experiment, the author observed (1) the activity of Na+-K+-ATPase, Ca2+-ATPase, and Mg2+-ATPase in rat brain tissues decreased significantly after 24 hours of injury (P <0.05), and the activity became even greater at 72 hours. (3) The activity of Na+-K+-ATPase in the 24-hour drug group was not significantly different from that of the model group (P> 0.05), and the activity of Mg2+-ATPase was not significantly different from that of the sham operation group (P> 0.05), but there is no significant difference compared with the model group (P>0.05), and the activity of Ca2+ -ATPase is significantly different from the model group (P<0.05). The activities of Na+-K+-ATPase and Ca2+-ATPase Mg 2+-ATPase in the 72-hour drug group were significantly different from the model group (P <0.05), and there was no significant difference compared with the sham operation group (P>0.05), it shows that the drug has a very good immediate response to regulating the activities of Mg 2+-ATPase and Ca2+-ATPase, and it can maintain a high level all the time. From the above, it can be explained that acupuncture can indeed keep the activity of the calcium pump and sodium pump at a higher level. Some scholars believe that there is a certain negative correlation between changes in Ca2+ -ATPase activity and changes in total calcium and that calcium accumulation in the early stages of brain injury is positively correlated with cerebral edema192. Some scholars believe that intracellular Ca2+ excretion is a factor in reducing intracellular Ca2+ overload1102. Acupuncture maintains high levels of calcium pump and sodium pump, which can further reduce intracellular calcium overload and cerebral edema, thereby protecting brain nerve cells. The mechanism may be that acupuncture can adjust the degree of vascular tension and cerebral perfusion pressure, improve cerebral blood supply and oxygen supply, provide energy for brain nerve cells, and delay their exhaustion and death. As for the acupuncture to regulate the activity of Na+-K+-ATP, Ca2+-ATPase, and Mg 2+-ATPase, it needs further study in the future.
