To study the protective effect of Total Glycosides of Cistanche on Alzheimer's disease (AD) model rats. Methods: Rats were injected with condensed Q amyloid (Ap25.35) into the dorsal part of the hippocampus using brain stereotactic technology to replicate the rat AD model. The experiment was divided into sham operation (is volume normal saline), model (iso volume normal saline), vitamin E (40 mg/kg), and Total Glycosides of Cistanche high, medium, and low dose (160.80.40 mg/kg) groups. The intragastric administration was started 4 days before copying the model, once a day for 14 consecutive days. Passive avoidance jumping experiment and electrical maze experiment were used to detect the learning and memory ability of rats; tissue homogenate and brain tissue slices of rat hippocampus were prepared to detect the activity of acetylcholinesterase (AchE) in hippocampus tissue of rats, and the rats were observed under electron microscope The intracellular C content in the hippocampal CA1 area. Results: Compared with the sham operation group, the rats in the model group had significantly longer reaction time and a significant increase in the number of errors (corpse VO.O1), hippocampal AchE activity was significantly enhanced (corpse V0.01), and intracellular C storage content in the hippocampal CA1 area increase. Compared with the model group, the reaction time of rats in the high, medium, and low dose groups of Total Glycosides of Cistanche was significantly shortened, the number of errors was significantly reduced (P<0.01), and the AchE activity in the hippocampus was significantly weakened (FV0.01 or household V0.05), The intracellular Cf+ content in the CA1 area of the hippocampus was significantly reduced. Conclusion: Total Glycosides of Cistanche can reduce the AchE activity and Ca2+ content in the hippocampus of AD model rats, maintain the normal level of acetylcholine (Ach) in the brain and improve the ability of learning and memory.

The previous work of this research group has confirmed that Total Glycosides of Cistanche have a significant protective effect on neuro cytotoxicity caused by A wound ball. 1.1 Instrument

1.2 Medicinal materials

Cistanche extracts (from Beijiang, Xinjiang) were identified as genuine by Chief Pharmacist Yang Jianhua of the Third Affiliated Hospital of Xinjiang Medical University.

1.3 Drugs and reagents

Total Glycosides of Cistanche (made by the Department of Natural Medicines/Biopharmaceuticals, School of Pharmacy, Xinjiang Medical University, the main ingredient is Phenylethanoild Glycosides, the purity determined by UV method is >50%); A Yuqiu (American Sigma Company, batch number: 115k4778); Vitamin E (Vit E), Shanghai Sanwei Pharmaceutical Co., Ltd., batch number: 20070605); AchE test kit (Nanjing Jiancheng Institute of Bioengineering).

1.4 Animals

60 adult healthy SD rats, 8, weighing (220±270) g, were provided by the Experimental Animal Center of Xinjiang Medical University (animal production certificate number: Yidongzi No. 16-003).

2 Method

2.1 Grouping and administration

The experiment is divided into 6 groups, namely sham operation (iso volume normal saline), model (iso volume normal saline), Vit E (50 mg/kg), and Total Glycosides of Cistanche high, medium, and low doses (160, 80, 40 mg) /kg) group, ig administration was started 4 days before copying the model, once a day for 14 consecutive days.

2.2 Copy of the model

Dissolve 1 mg of A wound ball in 200 J1 sterilized physiological saline, seal it and place it in a 37 T cell incubator, and form a condensed state after 7 days. Rats are anesthetized with chloral hydrate (400 mg/kg) and routinely prepared and disinfected. The skin is cut in the middle of the top of the skull, fixed with a stereotaxic instrument, and the head is flattened. Determined according to the stereotaxic map of the rat The backside of the right hippocampus is 3.8 mm, 2.5 mm next to the midline, and 3.0 mm below the surface of the skull, which is the dorsal part of the right hippocampus. Withdraw the cerebrospinal fluid, slowly inject within 5 minutes with a 10 microinjector, and retain the needle for 10 minutes. After penicillin sodium 50,000 u, once a day for 3 days.

2.3 Detection of indicators

2.3.1 Rats’ passive avoidance platform jumping experiment 14 days after the operation, but the rats into the platform reaction box, adapt to the environment on the copper grid for 3 minutes, turn on the power (90V alternating current), the rats received an electric shock, the normal reaction is Jump on the platform to avoid noxious stimuli.

Total Glycosides of Cistanche significantly improve the learning and memory ability of AD-induced rats, shortens the response time to electrical stimulation, reduces the number of errors, and reduces the activity of AchE and the accumulation of Ca" in the hippocampal CA1 area. In summary, Total Glycosides of Cistanche has a protective effect on the learning and memory impairment of AD model rats caused by A", and thus plays a role in the treatment of memory and cognitive impairment. One of its mechanisms may be to maintain the normal level of Ach in the brain and reduce the content of Ca. It is related to the learning and memory ability of AD model rats. Our group intends to further study from molecular biology whether Total Glycosides of Cistanche can inhibit the hyperphosphorylation of tau-protein deposition by down-regulating the activity of glycogen synthase kinase 30, thereby Inhibit the deposition of A and e, exerting its neuron protective effect.
